Background
| Alternative Names | C-reactive protein, CRP, Pentraxin-1, PTX1 |
| Function | C-reactive protein (CRP), also known as Pentraxin-1, is a key acute-phase protein that plays a role in the innate immune response. CRP acts as a pattern recognition molecule that binds to specific molecular patterns, such as phosphocholine residues found on the surfaces of various pathogens (e.g., bacteria) and damaged host cells. This binding facilitates opsonization, marking targets for clearance by phagocytic cells. CRP interacts with the C1q component of the classical complement pathway. This triggers a cascade leading to the formation of membrane attack complexes and further opsonization, enhancing pathogen elimination. CRP modulates the inflammatory response. It influences cytokine production, affecting the balance between pro-inflammatory and anti-inflammatory signals depending on the context and the microenvironment. CRP exerts many of its immunomodulatory effects through binding to Fcγ receptors (Fc gamma RI, Fc gamma RIIA, and Fc gamma RIIB) on dendritic cells (DCs) and macrophages. Engagement of Fc gamma receptors activates pathways such as the MAP kinase cascade and NF-kappa B signaling. These cascades lead to the transcription of pro-inflammatory cytokines. |
| Tissue Specificity | C-reactive protein (CRP) is primarily reported as a liver-derived acute-phase protein, where hepatocytes respond to pro-inflammatory cytokines (IL-6, IL-1 beta) to upregulate CRP expression. Lower levels and context-dependent expression of CRP is reported in adipose tissue, vascular cells (endothelial cells, smooth muscle cells), immune cells (macrophages and monocytes), and lung tissue. |
| Cellular Localization | Secreted |
| Involvement in Disease | Dysregulation of CRP is associated with cardiovascular and metabolic disorders (atherosclerosis, coronary artery disease, obesity, type 2 diabetes), rheumatoid arthritis, systemic lupus erythematosus (SLE), acute bacterial infections and sepsis. |
