Background
Alternative Names | Interleukin 1 alpha, IL-1A, IL1F1, Hematopoietin-1 |
Function | IL-1 alpha is a proinflammatory cytokine that plays a central role in regulating the immune response and inflammation. It is involved in the initiation and amplification of the inflammatory process by promoting the production of other cytokines, chemokines, and acute-phase proteins. IL-1 alpha activates immune cells such as macrophages, neutrophils, and dendritic cells, and contributes to the recruitment of these cells to sites of infection or injury. It is also critical in processes like fever induction, tissue repair, and the regulation of immune responses during infections, autoimmune diseases, and inflammatory conditions. IL-1 alpha is a glycoprotein with a molecular weight of approximately 17 kDa. It is initially synthesized as an inactive precursor (pro-IL-1 alpha) and is primarily expressed in the cytoplasm of epithelial cells, endothelial cells, and macrophages. IL-1 alpha exerts its effects by binding to the IL-1 receptor type 1 (IL-1R1). Upon binding, the receptor undergoes a conformational change, leading to the recruitment of adaptor proteins such as MyD88 (myeloid differentiation primary response gene 88) to the receptor complex. This initiates a signaling cascade involving the activation of the NF-κB pathway and mitogen-activated protein kinases (MAPKs). |
Tissue Specificity | IL-1 alpha expression has been reported in epithelial cells (skin, gastrointestinal tract, and respiratory system), endothelial cells, macrophages, fibroblasts, dendritic cells, keratinocytes, and neutrophils. |
Cellular Localization | Secreted protein (primarily), membrane bound |
Involvement in Disease | Dysregulation of IL-1 alpha is associated with rheumatoid arthritis (RA), inflammatory bowel disease (IBD), atherosclerosis, type 2 diabetes, cancer (inflammatory tumor microenvironment), psoriasis, Alzheimer’s disease, systemic lupus erythematosus (SLE), chronic obstructive pulmonary disease (COPD), and periodontal disease. |
