Background
Alternative Names | Interleukin-17F, ML-1, CANDF6 |
Function | Interleukin 17F (IL-17F) is a pro-inflammatory cytokine that belongs to the IL-17 family, which plays a significant role in immune responses, particularly in inflammation and tissue homeostasis. IL-17F is primarily involved in the defense against bacterial and fungal infections by promoting the recruitment of neutrophils and the production of other inflammatory cytokines. It also enhances the production of antimicrobial peptides in epithelial cells and induces the expression of inflammatory mediators such as IL-6, IL-8, and TNF-α. While IL-17F shares functional similarities with its family member IL-17A, it has a less potent effect on inducing inflammation and is thought to be involved in mucosal immunity, particularly in respiratory and gastrointestinal tissues. The receptor for IL-17F is primarily the IL-17 receptor (IL-17R) complex, which consists of several subunits: IL-17RA, IL-17RC, and other associated co-receptors. Upon binding of IL-17F to the IL-17RA/RC heterodimer, downstream signaling is initiated via the recruitment of adaptor proteins such as Act1 (also known as CIKS). Act1 activates the NF-κB, MAPK, and C/EBP signaling pathways. |
Tissue Specificity | IL-17F expression has been reported in lung tissue, intestinal tissue, skin, synovial tissue, CD4+ T cells, specifically Th17 cells, neutrophils, respiratory and gastrointestinal epithelium, macrophages, γδ T cells and innate lymphoid cells (ILCs). |
Cellular Localization | Secreted protein |
Involvement in Disease | Dysregulation of IL-17F has been implicated in autoimmune disease (rheumatoid arthritis, psoriasis, systemic lupus erythematosus), Crohn’s disease, ulcerative colitis, asthma, multiple sclerosis, chronic obstructive pulmonary disease (COPD), Type 2 diabetes, and cancer (tumor microenvironment). |
