Background
Alternative Names | Interleukin 5, B-cell differentiation factor I, BCDF mu, BCGFII, Eosinophil differentiation factor, EDF, Eo-CSF, T-cell replacing factor, TRF, ILN |
Function | Interleukin-5 (IL-5) is a cytokine primarily involved in the regulation of eosinophil development, differentiation, activation, and survival. It is key in orchestrating immune responses, particularly in allergic reactions, asthma, and parasitic infections. IL-5 plays a critical role in eosinophil production in the bone marrow and their subsequent release into the peripheral blood, where they accumulate at sites of inflammation. Additionally, IL-5 promotes the activation of eosinophils, enhancing their ability to release pro-inflammatory mediators. IL-5 is a glycoprotein that belongs to the IL-5/IL-3/GM-CSF cytokine family, sharing structural similarities with IL-3 and GM-CSF. IL-5 exerts its biological effects through binding to the IL-5 receptor (IL-5R), a heterodimeric receptor complex composed of two subunits: the IL-5Rα (alpha) subunit, which is specific for IL-5, and the common β (beta) chain shared with other cytokine receptors like IL-3 and GM-CSF. Upon IL-5 binding to the IL-5Rα, a conformational change occurs, leading to recruitment of the signaling component, the β chain. This activation triggers downstream signaling pathways, primarily involving the JAK/STAT pathway. |
Tissue Specificity | IL-5 expression has been reported in spleen and lung tissue, bone marrow, T-helper 2 (Th2) and Mast cells |
Cellular Localization | Secreted protein |
Involvement in Disease | Dysregulation of IL-5 is associated with asthma, particularly in eosinophilic asthma, eosinophilic esophagitis (EoE), allergic rhinitis, chronic rhinosinusitis with nasal polyps (CRSwNP), atopic dermatitis (eczema), parasitic infections, such as helminth, and celiac disease. |
